ERCC 1 ( excision repair complementing defective repair in Chinese hamster )
نویسنده
چکیده
ERCC1 is expressed at higher levels in tumor tissue compared to normal tissue and the expression shows more inter-individual variation among cancer patients than among healthy individuals. ERCC1 expression and ERCC1 protein levels in tumor tissue may predict response to chemotherapy. Thus, non-small cell lung cancer patients with undetectable ERCC1 protein levels in tumor tissue had a longer survival after cisplatinbased adjuvant chemotherapy than patients with detectable ERCC1 protein levels. However, high ERCC1 protein levels were associated with increased survival among patients who were not treated with chemotherapy.

Human repair gene restores normal pattern of preferential DNA repair in repair defective CHO cells.
The pattern of preferential DNA repair of UV-induced pyrimidine dimers was studied in repair-deficient Chinese hamster ovary (CHO) cells transfected with the human excision repair gene, ERCC-1. Repair efficiency was measured in the active dihydrofolate reductase (DHFR) gene and in its flanking, non-transcribed sequences in three cell lines: Wild type CHO cells, a UV-sensitive excision deficient...
متن کاملComplementation Using Rodent and Human Cell Extracts*
Numerous rodent cell lines exist that have defects in nucleotide excision repair of DNA caused by alterations in genes that fall into 10 different complementation groups. The precise roles in the repair of these genes are unknown. We report here that extracts from Chinese hamster ovary cells of excision repairdefective complementation groups 1 and 3 are defective in DNA excision repair in a ...
متن کاملHamster Ovary Cells to DNA Damaging Agents Effect of ERCC-1 Overexpression on Sensitivity of Chinese
Previous studies of ERCC-1 gene expression levels in chronic lymphocytic leukemia and ovarian carcinoma tumor specimens indicated that increased gene expression may correlate with a lack of response to the alkylating agents, melphalan, and cu-diamminedichloroplatinum (II). In order to demonstrate direct involvement of the ERCC-1 protein in repair of melphalan lesions, the ERCC-1 defective Chine...
متن کاملCharacterization of genes and proteins involved in excision repair of human cells.
To extend our knowledge of the excision repair system in mammalian cells we have focussed on the isolation of genes and proteins involved in this process. For the purification and characterization of human repair proteins the microneedle injection assay technique is utilized. This system is based on the transient correction of the excision repair defect of xeroderma pigmentosum (XP) fibroblasts...
متن کاملFormation of a ternary complex by human XPA, ERCC1, and ERCC4(XPF) excision repair proteins.
The xeroderma pigmentosum complementation group A (XP-A) protein, XPA, has recently been expressed in Escherichia coli in a soluble and fully functional form. An affinity column was prepared by linking the XPA protein to a solid support. When HeLa cell-free extract capable of excision repair was applied to the column, > 99.9% of the proteins were in the flow-through. However, the flow-through f...
